Applications

DNA Sequencing

The Equator GX1 and GX 8 have been applied to various stages of sample preparation in DNA Sequencing:

  • Automated plating of bacteria for colony picking
The functionality of the control software on the Equator enables the spotting of transformed bacteria in a defined and uniform pattern across an agar plate. This enables optimum colony growth that results in an increase in the efficiency of colony picking.
  • PCR reaction set-up for DNA Template preparation
Addition of PCR master mix at microlitre and sub-microlitre volumes.
  • Rolling Circle Amplification (RCA)
For customers using the rolling circle amplification technique the Equator system is ideally suited to dispensing amplification enzyme (e.g. Templiphi®) in nanolitre or microlitre quantities.
  • Sequence reaction set-up.
The miniaturization of sequencing reactions can have substantial cost benefits. Sequencing volumes have been reduced by as much as 10X compared to typical volumes without compromising the quality of the sequence data.

The functionality of the Equator GX8 also provides for maximum flexibility in establishing the correct workflow specific to your laboratory . An example of this flexibility has been demonstrated at a customer site where the GX8 is used to add Sequencing mix to 384 well plates. In this instance the independent channel control of the GX8 enabled the customer to add forward mix with four dispense tips of the eight available and reverse mix with the other four in one single dispense step. This not only assisted in increasing throughput and efficiency but importantly addressed a major requirement from an informatics perspective in terms of desired sample layout and subsequent detection.
